Description
Villin (VIL1) is a tissue specific actin modifying protein associated with actin filaments in microvilli and the terminal web of epithelial cells. It is involved in regulating actin dynamics, cell morphology, the epithelial-mesenchymal transition, cell migration, and cell survival. Deregulation of this gene may contribute to tumorigenesis. Loss of expression of villin is seen in poorly differentiated colorectal carcinomas, particularly in microsatellite-unstable (MSI) tumors, and is linked to poor survival. In normal tissues, it is expressed on luminal membranes (microvilli) of the gastrointestinal epithelium, as well as in proximal convoluted tubules of the kidney and gallbladder. VIL1 is positive in pulmonary adenocarcinomas, colon adenocarcinomas, and renal cell carcinomas. References: FEBS Lett 2008 582(14):2128; Patnaik 2016; Arango 2012; Jaudah 2013

Specificity
Recognizes a protein of 95 kDa , which is identified as villin. It is a major constituent in the microvilli, which compose the brush border of epithelial cells forming absorptive surfaces of the intestinal and renal proximal tubular epithelia. Anti-Villin labels the brush border area in the gastrointestinal mucosal epithelium and urogenital tract. Among neoplasms, villin is predominantly expressed in tumors of colorectal origin. Antibody to villin is useful in identifying malignant cells from primary and metastatic colorectal carcinomas. This antibody also labels Merkel cells of the skin.

Usage
Immunohistology (Formalin-fixed - 1-2 ug/ml for 30 minutes at RT - Staining of formalin-fixed tissues requires boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes.) Optimal dilution for a specific application should be determined.
