THBD (Thrombomodulin; CD141) is an endothelial-specific type I membrane receptor that binds thrombin. Binding of THBD to thrombin leads to decreased fibrinogen-to-fibrin conversion and activation of the coagulation cascade. The THBD/thrombin complex catalyzes protein C activation, which increases inhibition of coagulation by inactivating cofactors Va and VIIIa. Mutations in this gene are implicated in inherited thrombophilia and recurrent pregnancy loss.
