RXR gamma, a NR2 Hepatocyte NF4-Like Receptor, has been shown to affect the regulation of thyroid hormone target genes in thyrotropes, cell-type differentiation in the pituitary, peripheral metabolism, eye development, differentiation of limb myoblasts, and development or activation of cholinergic neurons in nigrostriatal extrapyramidal pathways. Mice lacking the RXR gamma gene have elevated levels of serum T4, thyroid-stimulating hormone, increased metabolic rate and display locomotor deficiences. The mouse RXR gene has two known protein isoforms (RXR gamma1 and RXR gamma2), produced via alternative exon splicing and differential promoter utilization. The mouse RXR isoforms exhibit distinct tissue-restricted expression patterns: RXR gamma1 is enriched in the brain, whereas RXR gamma2 is expressed widely in cardiac and skeletal muscle.
