Description
GABRA4 (Gamma-aminobutyric acid receptor subunit alpha-4) is a neurotransmitter receptor that binds gamma-aminobutyric acid (GABA) and is expressed in the brain. GABA is the primary inhibitory neurotransmitter in the central nervous system, causing a hyperpolarization of the membrane through the opening of a Cl- channel associated with the GABA-A receptor subtype. GABA-A receptors are important therapeutic targets for a range of sedative, anxiolytic, and hypnotic agents and are implicated in several diseases including epilepsy, anxiety, depression, and substance abuse. In immunohistochemistry, GABRA4 has highest membranous positivity in the cerebral cortex and basal ganglia in the brain, and is also found in other locatiosn in the central nervous system and at lower levels in heart muscle, the duodenum and the small intestine. Usage
Immunohistochemistry: LS-B3611 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3611 was determined to be 2.5 ug/ml. Peptide ELISA: antibody detection limit dilution 1:16000. Western Blot: Approx. 70kD band observed in Human Brain (Cerebellum and Frontal Cortex) lysates (calculated MW of 61.6kD according to NP_000800.2). Recommended concentration: 0.1-0.3 ug/ml.
