Description
Thyroglobulin (TG) is a protein synthesized by follicular cells of the thyroid, and is the precursor for thyroid hormones T3 and T4. Serum thyroglobulin measurements can be used as a marker for thyroid tumors, particularly in the postoperative management period, as well as a sensitive biomarker of iodine status in iodine deficiency states. Mutations in this gene are implicated in thyroid dyshormonogenesis with goiter, and congenital hypothyroidism. Polymorphisms are associated with autoimmune thyroid diseases such as Grave’s and Hashimoto’s thyroiditis. Immunogen
Recombinant full-length human Thyroglobulin protein. Antigen Molecular Weight: 660 kDa (Dimeric Form)
Specificity
Thyroglobulin is a 660 kDa dimeric pre-protein with multiple glycosylation sites. It is produced by and processed within the thyroid gland to produce the hormone thyroxine and triiodothyronine.  Prior to forming dimers, thyroglobulin monomers undergo conformational maturation in the endoplasmic reticulation.  The vast majority of follicular carcinomas of the thyroid will give positive immunoreactivity for anti-thyroglobulin even though sometimes only focally. Poorly differentiated carcinomas of the thyroid are frequently anti-thyroglobulin negative. Adenocarcinomas of other-than-thyroid origin do not react with this antibody. This antibody is useful in identification of thyroid carcinoma of the papillary and follicular types. Presence of thyroglobulin in metastatic lesions establishes the thyroid origin of tumor. Anti-thyroglobulin, combined with anti-calcitonin, can identify medullary carcinomas of the thyroid. Furthermore, anti-thyroglobulin, combined with anti-TTF1, can be a reliable marker to differentiate between primary thyroid and lung neoplasms.

Usage
Immunohistology (Formalin-fixed - 0.5-1.0 ug/ml for 30 min at RT - Staining of formalin-fixed tissues requires boiling tissue sections in 10 mM citrate buffer, pH 6.0, for 10-20 min followed by cooling at RT for 20 minutes.) Optimal dilution for a specific application should be determined.
